is your AV locking the file or there is some vm stuff taking a snapshot of the disk and freezing the i/o for some time.

i can connect from SQL 2000 QA to a SQL 2008 instance with out any issue. From enterprise manager you cannot connect to SQL 2005 or 2008

You can alternatively use BCP to export the data , Zip it (7za is free and also run from command line)
then using ftp you can copy to destination , unzip...
