I would get a test plan together using a test cluster (if you have one) and include in it;
Rebooting nodes (single & multiple) / removing the witness
Disabling HBAs
Unpresenting disks
Disabling NICs...

Just a suggestion as well. The original post doesn't mention anything about the need to automatically detect the names of over 100 servers..

I don't think you can change them in SSMS.. try deleting the current endpoints and re-creating them using the CREATE ENDPOINT command, specifying the port no. you want to use.

I've seen this problem before and I'm pretty sure SQL 2008 will need to use a different Listening Port no. because of the existing 2005 mirror.

Check the permissions on the drive, and if it still fails, try creating a test folder and backup to there.
