All editions other than Enterprise Edition supports up to 16 instances and Ent Edition supports upto 50 instances. This is on a stand alone Server, on a failover cluster Ent...

I second Lowell's view, the database backup might have been taken before those users existed in the database..

Do you see any sessions running against that database ? May be look at activity Monitor and filter by the database and see if there is any session connected...
