You might try looking at using terminal servers and using remote desktop to run the application at the "branches". It would be like in Windows XP with several users, except that many desktops can be running at once. DSL has a slower upload time and by running the application on a local server the data transfer to the application would be much faster. It would also give better security as you would only be transferring the screenshot of the application to the branches and not the data itself (which can be intercepted). This will also "centralize" your operation and allow you to set permissions as to who has what access, one location with the data secured, and you can also do remote control of the remote desktop to do "Help Desk". You can use DNS with Active directory to limit the access to server or you can set the server up to only accept password protected users. You don't have to worry about having to merge data from several locations as they will all be working with the same data. Depending on your business, this can give your employees more information to work with as they don't have to wait for data updates if records are changed in a different location. Also if you need to update your application, you only have to do it in one place, the terminal server, instead of on each employee computer. Each employee would have a shortcut on their remote desktop to run the application and the application would only need to be in one place.

I'm guessing you have the data source at each site so business can continue even if the connection goes down.
If you had the time and money, I would suggest rearchitecting the application to use Microsoft Sync Framework.

Can you run the DTS package throughout the day?
I agree with Trainer about using Terminal Services back to the main office. SoftGrid might be another option.
