That's quite a request you've made there - it is a fairly involved area with far too many variations and possibilities to reply in one (sensible) thread.
There are a number of resources available, but the best to start with is probably Gail Shaw's site - http://sqlinthewild.co.za/. Also articles by her at http://www.simple-talk.com/sql/performance/finding-the-causes-of-poor-performance-in-sql-server,-part-1/ and http://www.simple-talk.com/sql/performance/finding-the-causes-of-poor-performance-in-sql-server,-part-1/. They may also be on her own web site, but you can check that. You will see an inordinate number of posts on this site (as GilaMonster), so you can follow her postings here which are very informative.
Brent Ozar's site http://www.brentozar.com/ has a wealth of information too, as does Kimberly Tripp http://www.sqlskills.com/BLOGS/KIMBERLY/ and Paul Randall http://www.sqlskills.com/BLOGS/PAUL/
And of course, a huge number of discussions and articles on this site.
It looks quite intimidating, but all of these people are very approachable, provided you're willing to listen to their advice.
Have a look in these various areas and then by all means return with more specific questions - its the type of subject that you're better off dealing with in small 'chunks'. Its a big subject best digested in small pieces.
P.S. No offence meant to others on this site that have excellent articles for this area - but I don't want to swamp him with too many links. I'm sure he'll find you all eventually - I did. Quality tend to show through eventually.

Acquiring sufficient experience, knowledge and understanding of SQL Server and TSQL to be reasonably proficient at performance tuning takes at least as much time and effort as a university degree.

I also recommend starting with Gail Shaw's articles linked above. It's an excellent place to get started. I'd also suggest you pick up a copy of my book on exactly this topic. There's a link in my signature line right below here.
