  I've tried to setup a Maintenance Plan that will backup my DB daily. However, I have not had any luck with the backup job executing. I can backup the DB manually.

  I just noticed that the SQL Agent is stopped. Does this have any effect on the DB backup job?

  Yep.  SQL Agent handles all the scheduling within SQL.  It must be running for any automated job to execute via its schedule.
