  We have sql server 2005 installed in windows server 2008 R2. now we have plan to installing WIndows server 2008 R2 service pack . do we face any issue after installing this on SQl server side.

  Please advice.

  Are you installing on cluser node on stand alone?

  take a look at this, http://support.microsoft.com/kb/955725, clustering may be an issues as advised above but on the whole I don't see any issues
