SQL Server 2005 Express (no sp) is 53.5 MB
SQL Server 2005 Express SP2 is 36.5 MB

That's a diff in size of almost 20 MB. How can the SP2 be so much smaller?

The 36.5 MB file contains only the 32-bit version. The 53.5 MB file contains both the 32-bit and the 64-bit versions.

OK, that makes sense. Will the setup wizard automatically install the appropriate version i.e. 64-bit version on 64-bit systems and 32-bit version on 32-bit systems? Is it possible to run the 32-bit version on a 64-bit system?

Firstly, let me say that I don't have any 64-bit systems. In re-reading the first link [ http://www.microsoft.com/downloads/details.aspx?familyid=220549b5-0b07-4448-8848-dcc397514b41&displaylang=en ] I realized that there is no 64-bit version of SQL Server Express, only a 32-bit version that can run in a 32-bit WOW subsystem on 64-bit hardware/OS.
See Additional Information in the first link. An except follows:
That said, now I'm not sure what the difference is, except that the first link looks like the original release of SQL Server 2005 Express, while the second is Service Pack 2. I haven't tried to do a fresh install with the SP2 install file. If that is possible, I guess there is no reason to download the first file.
