Using SQL Server 2005 standard edition.
Will someone please explain when to choose 'append to the existing backup set' or 'overwrite all existing backup sets' for the Overwrite media backup option.
This is for a full backup.

Think of a backup set as essentially a file. You can overwrite, meaning essentially delete and recreate the file. Or you can append, which means add a second set of data to the file.
I don't know many people that still use backup sets. Most people tend to backup to a specific file, naming them with the timestamp.
