We (try) to do a mock move within the last 48 hours of going live with updates. THen you can script/automate the move fairly easily since only the changes that you are moving this time is in staging.
However, most of the time we have 5-6 projects going in the same database, which makes it more difficult. With about 30 developers I make them submit to me a list of which objects changed, along with the code to make the change. Then those scripts are QC'ed by me and run in the staging environment first.
All objects get checked into source safe as soon as they go into production, with a note of which project number they belonged to.
