What about permissions on the share (not just the actual directories).

Did you assign those permissions to the SQL service account?

1) check Both Instance should run on same (service) account.
2) Check backup location have full access.

Check the share "testing_backup". On the properties go to the share permissions tab and set permission for sql service accounts for both instances (if not the same).
Then go to security tab and set NTFS ACLs, again for both accounts.

Make sure that the SQL Agent service account also has permissions to the share..

Its a permission issue check both server SQL Server Agent rights level and share folder access level as well as domain.
