Not sure what you are asking. Do you want to be able to store the data in SQL Server and link from Access? yes, you can do that. Do you want to be able to link from SQL Server to Access? Yes, you can do that, too with a Linked Server.

You can import tables using the import wizard in SQL or the upsize wizard in access. Access queries will import as tables.

I have used http://www.microsoft.com/sql/solutions/migration/access/default.mspx for upgrading an MDB to a SQL 2005 database and an ADP. It worked well for me.

Where's the import wizard in SQL Server 2005?
Right-click on the database in SSMS object explorer, select TASKS --> IMPORT DATA ....
