Our business planners want to establish a secure Extranet site hosting SQL Server 2K which needs to communicate with another SQL Server 7.0 inside (ON NT 4.0 SP6 MACHINE ) via DTS. What
is an appropriate configuration?

Windows 2000 SP4. At the end of thus year, all support for NT 4 goes away. While the OS is fairly stable, there are still security issues being found and there will be NO patches. That alone should move you off NT4.
The configuration means that you need a firewall or router between the machines. And the secure setting should only allow the external SQL server to communicate with the internal server across a single port. It's a single rule to limit access. Other than that, be sure that you have secure passwords, if possible, don't allow SQL auth, change passwords regularly, don't use the SA account for access.
