my server is Win2008r2 and sql version is 2005 I want to run the DMA which I want to upgrade to SQL 2016, is it possible to run the DMA after installing the .NET framework 4.5 on WIN2008 r2

I would suggest setting up a virtual machine and trying it out. Even if someone says it's OK, you still need to verify it and test the scenario before performing the upgrade. One thing is for sure, SQL 2016 cannot be installed on your Win2008 R2 box.

As per Hardware and Software Requirements:
SQL Server 2016 (13.x) RC1 and later require .NET Framework 4.6 for the Database Engine, Master Data Services, or Replication. SQL Server setup automatically installs .NET Framework.
You can't run 2016 on .Net 4.5.
As for operating support (OS Compatibility), the oldest OS you can use is Windows Server 2012/Windows 8.
