Hi All,
Currently, I have my SQL Server 2005 installed on the D drive of the server. My question is if i change the drive letter, what impact will it have?
Will i need to re-install?

Do you mean you have the SQL application installed on D:\ or just the database .mdf and .ldf?

Just the SQL Application itself.

I would avoid it in this case unless you are prepared to move system databases and the resource database.

What about if i have seperate drives for Tempdb(F) and SQL Backups(H) on a cluster and i want to change their letters to T and B respectively, they dont have anything apart from this inside it. I feel that doing this doesn't have any risks, but still want to know your view.

Changing drive locations for Backups and tempdb is a lot less risky and more straight forward. Make sure you change your backup jobs for the new drive letter.
There isn't much concern around changing those. I might avoid B though since it is historically reserved for floppy drives (not present on the server I am sure - but best to avoid the possibility).
