can i use differential back up to initialize my transactional subscription?
my full back is too large to setup the replication for every maintenance so if i can use differential back up on top of my existing backup on subscriber.
any help appreciated.

Just to be clear i am thinking below steps
remove subscription
take differential back up on publisher
create subscription on the basis of differential back up above
restore differentail backup of publisher on subscriber(this step i am not sure how it will match the LSN)

A differential backup only contains the pages that have changed since the last full backup. This is why it is so much smaller than your full backup.
You will not be able to a differential backup because it will not have all of your database in it (unless every single page was updated, of course).

thnx , the only question here , is there any way i can restore publisher differential backup on top of my already existing subscriber database?

After you restore the Full back to your subscriber, you can then restore the diff on the subscriber.
