Our DR plan consists of full backups of the system database nightly. If we lost the data drive would there be any issues restoring the system databases from tape? Does the instance have to be up and running?

Yes I the instance needs to be up and running as there is nothing to connect to, if there is no instance. See How To: Restore the master Database in BOL for special instructions on restoring master.
msdb and model can be restored like any user database although you only need to restore model if you made changes to it.

You can't restore any database including system if the sql is not running...
You need to start the sql in single user mode restore master...
make you installed the same as before other wise you can't restore system dbs on to different build...
