I'm considering multiple instances of SQL on a single server to address a load testing environment. In the 2nd instance, do I need to change the default TCP port so that the connecting application knows how to connect to its database?

You need to start SQLBrowser service for your applications to be able to find and use the instance.
(unless you connect directly using the actual port number)

I understand the principals of SQL Browser, but we haven't utilized it yet. Do I just configure it to automatically start, or do we need to perform some configuration of it, or enhance the applications to utilize it?

You just need to start it. No configuration needed.
Just be sure you use Local System service account !
(just as a best practise. In clustered env. you need it to be local system)
