I have created SQL 2005 in development as another insatance of SQL server. I have only one database and One DTS.
I took the backup from 2000 and restored in 2005. How should I migrate the DTS in 2005.
Can someone please help me

You cannot migrate DTS packages. You can only save them as structured files and run them from within SSIS. See BOL for details but you'll be better off rewriting the package in SSIS.
Regarding the DB upgrade: Did you change the compatibility to 90 and for good measure, change the Page Verify property to CHECKSUM?
